The Miller Laboratory at the University of Virginia School of Medicine seeks a research scientist with a background in molecular biology or genetics to apply functional genomic analyses to investigate inherited vascular diseases and cardiomyopathies. Available projects include the application of single-nucleus (e.g., snATAC/snRNA-seq, snCUT&Tag) and spatial (spatial transcriptomics, proteomics) genomic analyses of healthy and diseased subjects of ancestrally diverse backgrounds. The laboratory is interested in characterizing the cellular, molecular, and genetic mechanisms and candidate drug targets for subtypes of cardiovascular diseases with high unmet clinical need. <h3>Some of the specific goals of this work are to</h3> <ul> <li> Optimize scalable single-nucleus and spatial genomic technologies for fresh and archived human biospecimens. </li> <li> Apply targeted and systematic genome engineering approaches to perturb candidate regulatory elements or genes in relevant cellular models. </li> <li> Work with collaborators to develop and optimize phenotypic screens for genetic and drug perturbations. There will also be opportunities to develop bioinformatics and computational skills through collaborations on multiple internal and external projects. </li> </ul> The Research Scientist will have access to a broad range of cutting edge wet-lab and computing resources, and will be exposed to a stimulating, multi-disciplinary and inclusive environment in the Center for Public Health Genomics (CPHG). This work will involve close collaborations with other CPHG labs to broaden training in both wet lab and computational biology skills. A position is available at the Beirne Carter Center for Immunology Research ( CIC ), at the University of Virginia in the research group of Dr. Coleen McNamarato study the role of IgE to Alpha-gal in atherosclerosis. We have shown that humans with IgE to Alpha-gal have increased coronary artery plaque volume and features of plaque instability, as measured by intravascular ultrasound (Arterioscler Thromb Vasc Biol. 2018 Jul;38(7):1665-1669). Follow on studies implicated the CCL20-CCR6 axis in B cell class switching to IgE in humans (Front Cardiovasc Med. 2022 Jan 13;8:791028). We are now seeking a deeper understanding of the fundamental mechanisms involved.
